Model answer (4 marks)

Pepsin is secreted by the stomach and functions best at an acidic pH of about 1.5, where it cleaves peptide bonds in proteins. Trypsin is released in the small intestine and works optimally at a neutral to slightly alkaline pH (≈7.5). Each enzyme would be denatured or inactive if it encountered the opposite pH; pepsin would lose activity in the intestine and trypsin would be inactivated in the stomach. Therefore, both enzymes are required to ensure proteins are progressively broken down into peptides and amino acids throughout the alimentary canal, achieving complete digestion.

Examiner tips

  • Mention the specific pH optima for each enzyme and the site of action; link pH to enzyme activity.
  • Explain that each enzyme would be denatured in the other's environment, justifying the need for both.
  • Show the sequential action – stomach then small intestine – to cover the whole digestive tract.

  2. Trypsin works in the small intestine where conditions are neutral or slightly alkaline, which is its optimum pH.
  3. Each enzyme would be denatured / stop working in the wrong pH / in the other enzyme's location.
  4. Having both enzymes ensures protein is broken down / digested to amino acids throughout the alimentary canal / completely.
